Analysis of diversity among six populations of Colombian mango (Mangifera indica L. cvar. Hilacha) using RAPDs markers Electron. J. Biotechnol.
Díaz-Matallana,Marcela; Schuler-García,Ingrid; Ruiz-García,Manuel; Hodson de Jaramillo,Elizabeth.
Currently in Colombia, there are only records of morph-agronomic characterizations of Mangifera indica cvar. Hilacha; molecular studies on this mango variety have not been carried out. The aim of this work was to identify the genetic diversity of six populations of mango Hilacha by RAPDs markers, as a fundamental base for breeding programs, conservation and selection of promissory materials for the fruit industry at the national level. From 60 primers evaluated in the populations, five primers were selected and were launched in the six populations. Polymorphic bands of RAPDs were transformed into binary matrices, which were then processed with NTSYS-PC, POPGENE and TFPGA softwares. The overall genetic diversity, H T = 0.468 ± 0.0016, is very similar to the...

Peteira,Belkis; Bernal Cabrera,A; Martínez,B; Ileana,Miranda.

Efficiency of RAPD and ISSR markers system in accessing genetic variation of rice bean (Vigna umbellata) landraces Electron. J. Biotechnol.
Muthusamy,Saraladevi; Kanagarajan,Selvaraju; Ponnusamy,Shanmugasundaram.
Vigna umbellata (Thunb.) Ohwi and Ohashi commonly known as rice bean or climbing mountain bean is under-exploited tropical legume. Genetic variation between 10 landraces of rice bean was evaluated using random amplified polymorphic DNA (RAPD) and inter simple sequence repeats (ISSR) markers. Among these markers, RAPD primers generated 987 amplification products of which 719 were polymorphic and ISSR markers produced 479 amplification products, out of which 296 were polymorphic. RAPD fingerprinting detected more polymorphic loci (70.30%) than the ISSR fingerprinting (61.79%). Mean PIC (polymorphism information content) for each of these marker systems (0.243 for RAPD and 0.203 for ISSR) suggested that both the marker systems were equally effective in...

Rabbani,Malik Ashiq; Pervaiz,Zahida Hassan; Masood,Muhammad Shahid.
The molecular marker is a useful tool for assessing genetic variations and resolving cultivar identities. Information on genetic diversity and relationships among rice genotypes from Pakistan is currently very limited. The objective of this study was to evaluate the genetic polymorphisms and identities of 10 traditional, 28 improved and 2 Japanese cultivars of rice using the random amplified polymorphic DNA technique. Twenty-five decamer-primers could generate a total of 208 RAPD fragments, of which 186 or 89.4% were polymorphic. The number of amplification products produced by each primer varied from 4 to 16 with an average of 8.3 bands primer-1. The size of amplified fragments were ranged from 200 to 4000 bp. Pair-wise Nei and Li's similarity had...

Morphological and genetic characteristics allow the identification of a collection of garlic cultivars in the North-Central region of Mexico Phyton
Azuara Hern ández,L; Silos Espino,H; Perales Segovia,C; Gómez Leyva,JF; Alpuche Solís,AG; Macias Valdez,LM.
The objective of this study was to compare the yield and genetic relationships between two Perla garlic selections, obtained by individual selection of cloves, and commercial varieties and cultivars adapted to diverse regions of Mexico (which did not have a selection process). Varieties with fewer cloves showed higher yields. Coreano and California produced 20 ton/ha (fresh weight) and Perla selections between 17-19 ton/ha (fresh weight). Six primers of the series OPB were tested for genetic characteriza-tion and OPB-17 was selected. With the amplified DNA fragments, a binary matrix was generated; afterwards, a dissimilarity matrix and dendrogram were developed. High genetic diversity was found among all varieties, which were separated into two groups...

Morphological and genetic differentiation among Chilean populations of Bufo spinulosus (Anura: Bufonidae) RChHN
Méndez,Marco A.; Soto,Eduardo R.; Correa,Claudio; Veloso,Alberto; Vergara,Eliseo; Sallaberry,Michel; Iturra,Patricia.
Bufo spinulosus has a wide and fragmented range distribution in Chile (18° to 33° S) along altitudinal and latitudinal gradients. Genetic variation was estimated using RAPD (Random Amplified Polymorphic DNA) markers in 10 populations from northern and central Chile. Morphometric and genetic information was analyzed as a function of geographical origin. The correlation between genetic and morphometric differentiation was analyzed by the Mantel test. An increase in body size as a function of latitude was observed. Specimens from El Tatio had the smallest body size and the greatest morphometric divergence. The AMOVA applied to genetic data indicated that 57.85 % of the variance is explained by interregional differences and that 30.12 % of the...

Phylogenetic relationships among species of the subsection Dendrophlomis Bentham Electron. J. Biotechnol.
Yuzbasioglu,Ertugrul; Dadandi,Mehmet Yasar.
This study used randomly amplified polymorphic DNA markers to determine genetic relationships among species of the subsection Dendrophlomis. Twenty accessions of the eleven Phlomis taxa were evaluated to determine genetic variability using fourteen ten mer primers selected from a 125 random oligonucleotide set. These 14 selected primers generated 85 RAPD bands that ranged in size from 200 to 1200 base pairs. Of the total bands, 88% (75) were polymorphic among the samples. Genetic distances among accessions were computed to produce a dendrogram based on UPGMA. Genetic distances ranged from 0.133 (between P. amanica and P. monocephala) to 0.494 (between P. chimerae and P. lunariifolia). The UPGMA tree based on distances has two major groups. The first...

Small-scale morphological and genetic differentiation in the Mediterranean killifish Aphanius fasciatus (Cyprinodontidae) from a coastal brackish-water pond and an adjacent pool in northern Sardinia ArchiMer
Maltagliati, F; Domenici, P; Fosch, Cf; Cossu, P; Casu, M; Castelli, A.
Two samples of Aphanius fasciatus collected in the Pilo pond (northern Sardinia, Italy) and in an adjacent pool of small surface area were analysed morphologically (235 individuals) and genetically (a subsample of 58 individuals). The aims of the present study were (i) to test the hypothesis that different predation pressures may be associated with morphological and/or genetic differences between samples from each habitat and (ii) to assess the level of divergence between the two populations. Morphological analysis was based on the relative size of fins because it has been shown to be associated with predation pressure. The relative caudal fin area (caudal fin area/total body surface) was smaller in specimens from the pool, in both males and females,...
